Abstract
[32P] γ-ATP와 [32P] α-dCTP는 대표적 P-32 표지화합물이며, 인산화 시약 또는 여러 RNA나 DNA의 P-32 표지 기질로 사용된다. 이 화합물들은 주로 생화학적 합성법에 의해 합성이 된다. 본 연구실에서는 P-32 표지화합물의 대량생산에 맞는 화학적 합성방법으로, 짝지움 시약인 O-8-(5-chloroquinolyl)-S-phenyl phosphorothiate을 이용하였다. O-8-(5-chloroquinolyl)-S-phenyl phosphorothiate의 총 합성 수율은 42%이었다. 이를 이용한 [32P] γ-ATP와 [32P] α-dCTP의 합성표지수율은 각각 95%, 40%였다.

[32P] γ-ATP (adenosine 5'-[γ-32P]triphosphate) and [32P] α-dCTP (deocycytidine 5'-[α-32P]triphosphate) are representatives of P-32 labeled compounds and used as phosphorylating agents or labeled substrates for several RNA and DNA polymerase reactions. These are usually produced by enzymatic processing. Our studies have been focused mainly on preparing these compounds by chemical processing for mass production. O-8-(5-chloroquinolyl)-S-phenyl phosphorothiate, a coupling agent, is proper for the systhesis of [32P] γ-ATP and [32P] α-dCTP. Coupling agent has been prepared in a yield of 42%. [32P] γ-ATP and [32P] α-dCTP were prepared in radiochemical yields of 95% and 40%; respectively.
