Synthesis of Hydroxyphosphonate Derivatives Via Mukaiyama Reaction

Mukaiyama 반응을 이용한 Hydroxyphosphonate 유도체 합성에 관한 연구

phosphonoaldehyde를 포함하는 Mukaiyama 반응을 처음으로 시도하였고, 반응의 최적화를 위해 diethyl phosphonoacetaldehyde와 1-trimethylsioxy-1-phenylethene의 반응을 모델반응으로 Lewis 산 및 용매를 변화시킨 결과 염화메틸렌과 BF3·OEt2에서 반응이 가장 잘 진행됨을 알 수 있었다. 반응의 다양성을 알아보기 위해 phosphonoaldehyde와 silyl nucleophile을 변화시킨 결과, 본 반응이 hydroxyphosphonate를 포함하는 다양한 유도체를 만들 수 있는 좋은 방법임을 증명하였다. 위의 방법으로 합성하여 얻은 새로운 화합물은 다음과 같다. Diethyl 4-oxo-2-hydroxy-4-phenylbutylphosphonate, diethyl 4-oxo-2-hydroxy-4-methoxybutylphosphonate, diethyl 2-hydroxypent-4-enylphosphonate, diethyl 2,2-trimethylsiloxycyanoethylphosphonate, diethyl 2-hydroxy-2-(2-oxocyclohexyl)ethylphosphonate, diethyl 3-hydroxy-3-(2-oxocyclohexyl)propylphosphonate. 이들 화합물은 1H NMR spectroscopy, IR spectroscopy 와 GC/MS로 확인하였다.

Mukaiyama reaction of silyl nucleophiles with phosphonoaldehydes was developed. In order to find the optimum condition, various Lewis acid, solvent were examined by taking the reaction of diethyl phosphonoacetaldehyde with silyl enol ether of acetophenone as a model and finally BF3·OEt2 in dichloromethane was found to be an excellent catalyst. This method was proved to be a good method to synthesize the hydroxyphosphonate derivatives via reaction of various silyl enol ether with phosphonoacetaldehyde. Previously unreported compounds were synthesized. They are diethyl 4-oxo-2-hydroxy-2-phenylbutylphosphonate, diethyl 4-oxo-2-hydroxy-4-methoxybutylphosphonate, diethyl 2-hydroxypent-4-enylphosphonate, diethyl 2,2-trimethylsiloxycyanoethylphosphonate, diethyl 2-hydroxy-2-(2-oxocyclohexyl)ethylphosphonate, diethyl 3-hydroxy-3-(2-oxocyclohexyl)propylphosphonate. All the compounds prepared in this study were identified by means of 1H NMR spectroscopy, IR spectroscopy and GC/MS spectrometry.
