Abstract
단일가닥구조다형성(single strand conformation polymorphism, SSCP)에 바탕을 둔 모세관전기영동법 (capillary electrophoresis, CE)을 이용하여 한우와 수입우의 빠르고 간단한 판별법을 개발하였다. 한우와 수입우 의 특정한 단일염기다형성(single nucleotide polymorphism, SNP) 표지인자를 중합효소연쇄반응을 통해 증폭하였 다. 증폭된 이중나선-유전자(ds-DNA)에 대해 SSCP 방법을 통해 단일가닥-유전자(ss-DNA) 단편들로 변환시킨 뒤, 2.2%(w/v) poly(ethyleneoxide)(Mr 600 000)와 1.0%(w/v) poly(ethyleneoxide)(Mr 8 000 000)의 혼합젤을 충진시킨 용융-실리카 모세관(75 μm 내경 × 365 μm 외경; 30 cm 유효길이; 50 cm 총길이)을 이용하여 분리하였다. SSCPCE 방법에 의해 360 V/cm의 전기장에서 한우와 수입우를 단지 13분 이내에 간단히 판별할 수 있었다.

We developed a rapid and simple method for distinguish of Hanwoo and imported beef by capillary electrophoresis based on the single strand conformation polymorphism(SSCP-CE). PCR amplification was performed to amplify specific DNA regions spanning single nucleotide polymorphism(SNP) markers to determine between Hanwoo and imported beef. The SSCP ss-DNA fragments from the amplified PCR products were analyzed through the fusedsilica capillary(75 μm I.D. × 365 μm O.D.; effective length, 30 cm; total length, 50 cm) filled with the mixed polymer of 2.2%(w/v) poly(ethyleneoxide)(Mr 600 000) and 1.0%(w/v) poly(ethyleneoxide)(Mr 8 000 000) as a sieving matrix. Under the electric fields of 360 V/cm, Hanwoo and imported beef were simply distinguished within only 13 min by SSCP-CE.
