Extraction and Purification of Eupatilin from Artemisia Princeps PAMPAN

사자발쑥에 포함된 유파티린의 추출 및 정제

쑥의 추출물은 항염작용, 항암 등 약용으로 많이 사용되고 있다. 본 실험에서는 제조용 크로마토그래피를 이용하여 사자발쑥에 포함되어 있는 유파티린을 추출하고 분리하기 위해서 최적조업조건을 실험적으로 구하였다. 사자발쑥을 에탄올로 추출하고 헥산, 클로로포름, 에틸아세테이트로 각각 분배하여 용매추출을 하였다. 액체 크로마토그래피의 이동상 조성과 주입 부피는 각각 water/acetonitrile/TFA=50/50/0.5(vol.%), 20 ml 이었다. 유속은 l.0 ml/min이며 UV wavelength는 370 nm 이었다. 유파티린이 함유된 피크를 제조용과 분석용 컬럼을 이용하여 분취하였고 LC-CE-MS로 정성분석을 하여 유파티린을 확인하였다.

The extracts of the whole herb of Artemisia princeps PAMPAN have been used for the treatment of inflammation, cancer and other disorders. In this experiment to extract and separate eupatilin contained in Artemisia princeps PAMPAN using a preparative chromatography, the optimum operating conditions were experimentally determined. eupatilin was extracted from the leaf and trunk of Artemisia princeps PAMPAN by ethanol. The resulting solution was further partitioned with n-hexane, chloroform and ethyl acetate. The mobile phase composition and the injection volume were water/ acetonitrile/TFA=50/50/0.5 (vol. %) and 20 ml, respectively. The flow rate was fixed at l.0 ml/min and UV wavelength was 370 nm. The measured retention time of eupatilin was approximately 8.6 min. The solution which contained eupatilin was collected by preparative and analytical column, and was identificated by LC-CE-MS.
